No, a guy losing his virginity doesn't hurt.
It can hurt to do missionary, depending on the area you're doing it in, sore knees, carpet rash or if you are supporting your weight on your arms they and your wrists can get sore too. If you have any injuries (back pain etc) it can make that worse too.
I doubt it would hurt. We're not the ones getting stretched like some laffy taffy.

it wont hurt the man butthe woman it is possible. if he goes in too aggressively, or she is not ready, he might hurt her. also the woman usually will bleed a small bit
