The 2 handed overhead throw incorporates a lot of muscles that "activate" and "deactivate" at different points in the movement.

Off hand I can name a few extremely important muscles. The most important would be the Rotator cuffs which are a group of muscles (subscapularis, supraspinatous, infraspinatus and teres minor). Without these the shoulder would literally not function properly. I'd also say the Anterior Deltoid along with the Medial and possibly the Posterior.

Thats just the shoulder.

You would also make use of the Latissimus Dorsi to bring the shoulder forward.

And in the arm the Triceps would lay a key role in extending the arm.

You would also use the flexor and extensor muscles in the forearm to grip and let go of the object.
