You usually floss first and then brush. Brush at least twice a day if not three times. If you can't floss before each brushing, floss before going to bed.

I always floss at night before bed and just before brushing. I like to think my teeth do not have food stuck in between them while I am sleeping. brushing 2 times a day is enough as doing so more often can damage your gums and slowly brush away the enamel on your teeth. Believe me I know.
