Yes, both can if you get allergies or sinitus, or an infection, but don't worry, you can always breath through your mouth. Now, usually, on side is plugged due to plain old gravity. Just switch sides while laying down for a while and you will feel the plugged side starting to loosen up.

Open a window or go outside and feel the blockage of your nostrils start to clear almost immediately.
Similarly it can happen when you are asleep, making you snore or even wake up in a panic.
Again, open a window, turn off any heating whilst you sleep, and an extra pillow might help.
