Not unless you breath in only helium for extended periods of time.
Doing that will cause oxygen deprivation and asphyxiation.
One balloon won't do anything but make your voice high pitched.

It can be harmful full strength and possibly deadly. However scuba divers use gas mixtures that include helium. So, I would not recommend doing this on a regular basis but it is not deadly in small doses.
