No, it doesn't kill all the sperm right away.
It helps, but most of the ejaculate isn't sperm at all: it's seminal fluid, which helps coat and protect the sperm. Spermicide can kill any sperm it reaches, but there are a lot of sperm swimming around in that seminal fluid which never even touch the condom itself.
This is why you need to change a condom right away after every single ejaculation. If semen leaks out, then there's a very good chance that the sperm isn't dead.
