The 'mini' pill and the drug amitriptylline do not interact. I am a little concerned as to why you are on amitriptylline however. It is no longer used as an antidepressant, though it is still used as a migraine prophylactic and also as a mood stabiliser. If it is being legally prescribed, your prescriber had a duty to confirm there was no interaction and advise you accordingly.

No, unless the amitriptylene makes you vomit, which could interrupt the pill's absorption into your body. Amitriptylene is a mild antidepressant. The only drugs that have been shown to make the pill "stop working" are antibiotics, and even then only one specific antibiotic ever showed statistically significant evidence that it could counteract the effects of the Pill. It was called rifampin, and it has fallen out of use because it was only used to treat tuberculosis.
You should always use back-up protection when taking antibiotics just to be on the safe side, but other drugs making the Pill stop working is more or less a medical myth. Some drugs may decrease the effectiveness, or the Pill could increase or decrease the effectiveness of other drugs. If you're really really concerned, just ring up your gyno and ask her.
