To make them really strong and defined when i wear shorts?

Yes! Bicycling is great for muscle definition for the whole leg. But you need to make sure you're challenging yourself with inclines, etc.
Also, if you've got a thick layer of fat over them, no amount of muscle will look defined. If you really want definition, you've got to work aerobically to reduce your body fat percentage.
